We investigated the TP53 gene in 44 glioblastoma (GBM) specimens from Chinese patients. Mutant p53 protein was detected in 40 of 44 (91%) specimens and in 69 of 94 (73%) cases using protein-based assays (EMD Laboratories). The incidence of mutations in the GBM specimens was 98%. Seventy-six percent (30/40) of the mutant tumors harbored a single point mutation in exon 5-8. All of the tumors with exon 4 mutation were located in the brainstem region. Truncating mutations were detected more frequently (30/40, 75%) than missense mutations (10/40, 25%). The screening of the entire TP53 gene identified 29 mutations in 14 patients, involving 15 cases in exon 5-8, exon 11, exon 6-9, and exon 10, but not exon 4 or exon 2-3. Three additional patients exhibited mutations at the same amino acid sites (located at codons 157 and 165) as patients with exon 4 mutations, but more than one mutation was detected in one case. The mutations were clustered in exons 6-9 and 11 in two cases. In addition, two mutational hot spots were identified, involving codon 152 and codon 165. All of these cases exhibited wild-type TP53 protein by immunohistochemical analysis, and functional analysis of the exon 9 and 10 mutations demonstrated that the mutant proteins were functional in suppressing transcriptional activation of the p53 target genes, but not in inducing apoptosis. On March 14, I attended the NAPAP Daylong Seminars. I learned about the varying legal issues that impact designers including architectural, site work, master planning, and urban design. I learned about the Environmental Advisory Council and how they interact with the APA. I learned a lot.

After the NAPAP conference, I was excited to join the Committee of Standards and to work with future nurses.
